I'm looking at getting a 3rd generation Sienna and trying to understand if there are differences between the main (or only) two middle row seats on the 7- and 8-passenger models (other than when the center "seat" is installed).

Yes they are conpletely different. The 2 vans also have different seat tracks in the floor as well so the seats are not interchangeable. The 8 passenger seat on the driver-side is much wider to accommodate the middle seat and hence has a different set of tracks on the floor of the van. The floorboard of the vans are different also because if this.
